Confocal imaging can benefit almost all researchers who currently use fluorescence microscopy. Confocal microscopy allows samples to be optically sectioned, rather than physically sectioned; greatly decreases the out-of-focus fluorescent emission; and permits three-dimensional imaging and data analysis. Our setup comprises a CARV Confocal Imaging System with filters (see below for a list of available filters), high-sensitivity Photometrics digital camera, and Metamorph imaging software, attached to a Nikon Eclipse E600 Research Microscope with Plan Fluor Phase 10x, 20x, 40x DIC objectives, Plan Apo DIC 40x objective and a Plan Apo DIC 60x oil objective. The Photometrics CoolSNAP fx camera is a fast, high-resolution, cooled CCD digital camera system designed for low-light scientific and industrial applications. It provides 12-bit digitization at 20 MHz. The fine pitch of the pixels, 6.7 x 6.7 microns, is ideally matched to the resolution of optical microscopes. Megapixel resolution and small pixels allow imaging of very fine detail, yet the pixels can be easily binned to improve sensitivity. Since this system uses a standard mercury light source, fluorochromes of any wavelength may be visualized by just buying the correct filter set.
